To study the effect of urolithin A on cytokine production in multiple sclerosis (MS) patients and healthy donors. The effect of urolithin A on stimulated production of interleukin (IL)-6 and interleukin-10 by peripheral blood mononuclear cells (PBMCs) and macrophages in patients with MS and healthy donors was evaluated using enzyme immunoassay. Basal and lipopolysaccharide-stimulated production of IL-10 PBMCs in patients with MS is lower than in healthy donors. In contrast, basal and stimulated production of IL-6 by PBMCs is higher in patients with MS compared to healthy donors. Urolithin A at 5 μM contributed to an increase in the production of IL-10 by PBMCs both in MS patients (by an average of 28%) and in healthy donors. Urolithin A also caused a decrease in the macrophage production of IL-6 in healthy donors (by an average of 10%) and MS patients. Given the beneficial immunotropic effect of urolithin A, this metabolite of the intestinal microbiota can be considered a promising compound for further study in patients with MS, including in clinical placebo-controlled studies.
