The nature of the biofilm adhering to pumice in an anaerobic filter treating molasses effluent has been studied by scanning electron microscopy (SEM). Samples were prepared for SEM using a variety of techniques, some of which are shown to produce artefacts interpreted by other authors to reflect the state of the biofilm. The formation of fibrillar structures is such an artefact of the sample preparation. Samples prepared using the Sputter-Cryo technique showed the pumice to be covered by a slime which prevents visualisation of the bacteria. This slime could be disrupted by preparative SEM techniques involving fixation and drying to produce a fibrillar network through which the bacterial consortium could be seen. Most of the slime and many bacteria were removed from the pumice during critical point drying. It is concluded that the Sputter-Cryo technique preserves the integrity of the biofilm while other methods can be utilised for visualisation of the bacteria.
