Pentafluorosulfanylation (-SF5) has become an increasingly valuable strategy in modern synthetic chemistry because of the distinct electronic and lipophilic properties of the pentafluorosulfanyl group. Recognized as a bioisostere of the -CF3 and tBu groups, it has attracted considerable interest in pharmaceutical, agrochemical, and materials science applications. Recent progress in pentafluorosulfanylation has led to the development of innovative approaches, facilitating the efficient incorporation of -SF5 into diverse molecular frameworks. This review explores key advancements while highlighting emerging applications. The continuous evolution of -SF5 chemistry is expected to drive further innovation in drug discovery, functional materials, and beyond.
