Optimization of extraction and macroporous resin purification processes of total triterpenoid from Boletus edulis Bull.: Fr.

Total triterpenoids are a significant bioactive substance in Boletus edulis; however, suitable extract and purification method used to prepare total triterpenoid has rarely been reported. In this study, the total triterpenoid from Boletus edulis was extracted, and the optimal extract parameter was determined by response surface methodology. The optimal extraction parameters were as follows: liquid–solid ratio of 20 ml/g, ethanol concentration of 70%, extraction temperature of 70°C, and extraction time of 60 min. Under these conditions, the extraction yield of total triterpenoid was 1.85%. The crude extract was further purified by AB-8 macroporous resin. The optimum adsorption conditions were: sample concentration of 3.5 mg/ml, pH of 5, and adsorption flow rate of 1.5 ml/min. The optimum desorption conditions were: ethanol concentration of 70%, and desorption flow rate of 1.0 ml/min. After purification, the content of total triterpenoid reached 8.71%, which was 2.49 times higher than that before purification. Novelty impact statement Studies on the extraction and purification of total triterpenoid from macrofungi have important research value. Up to now, there has been little information on the extraction and purification technologies to prepare total triterpenoid from Boletus edulis. Therefore, this study lays a theoretical foundation for the industrial production of total triterpenoid and also facilitates the further utilization of total triterpenoid from Boletus edulis as a functional food and drug.
