Influences of ferulate acid, indomethacin, indomethacinum-ferulate acid and imdomethacinum-ferulate acid-starch on rabbit platelet aggregation and production of thromboxan B_2 and 6-keto-prostaglandin F_(Iα) in vitro

objective To investigate the effect of indomethacinum-ferulate acid (FI) and indomethacinum-ferulate acid-starch (FIS) on rabbit platelet aggregation induced by ADP and AA and on the production of thromboxane B2 (TXB2) and 6-keto-prostaglandin F_(Ⅰα)(6-keto-PGF_(Ⅰα)) Methods Turbimetry and radioimmunoassay were employed to measure in vitro the platelet aggregation and the content of TXB_2, 6-keto-PGF_(Ⅰα) production in rabbit blood treated with ferulate acid (F), indomethacin (I), FI and FIS respectively. Results It was shown that F, I, FI and FIS significantly inhibited ADP-induced rabbit platelet aggregation and the production of TXB_2 and 6-keto-PGF_(Ⅰα) With the dosage of 12.5 to 240.0 μmol/L, the inhibition rate (%) of platelet aggregation by F was 11.3%-31.8% (ADP-induced) and 3.3%-18.5%(AA-induced), that by I was 5.3%-34.3% (ADP-induced) and 6.2%-20.2% (AA-induced), that by FI was 25.0%-59.3% (ADP-induced) and 30.3%-63.1% (AA-induced), and that by FIS was 13.0%-39.1% (ADP-induced) and 11.5%-39.6%(AA-induced) With the dosage of 240.0 μmol/L, the inhibition rates of TXB_2 and 6-keto-PGF_(Ⅰα) production by F was 14.8% and 1.2%.that by I was 24.9% and 26.4%. that by FI was 40.6% and 29.3% and that by FIS was 28.7% and 21.4%.
