AIM To improve the method of isolating single myocardial cells and to measure the basic electrophysiological properties. METHODS The improved Langendorff perfusing method and the patch clamp whole cell recording were used. RESULTS The resting membrane potential and whole cell currents was -85±2.3 mV ( n =10). The typical whole cell calcium currents could be recorded. CONCLUSION The isolation method can produce large numbers of single calcium tolerant myocardial cells with normal electrophysiological properties.
