Objective To develop a method for extracting and determinating polysaccharides from cultured Cordyceps militaris L.Link.Methods Orthogonal experiment was used to confirm the way of extracting.Contents of polysaccharides were detected by using phenol-sulphate acid.Results The best condition for extracting polysaccharides was by 80 times of pure water,once at 45 ℃,10 min for one time.Linear range was 10~100 g·mL-1,r = 0.999 8,the recovery rate was 99.41%,RSD= 2.36%.The average concentration of polysaccharides in three samples was 22.12%.Conclusion The extracting method is simple,reproducible and the results is accurate and reliable.It can be used to extract and determine the contents of polysaccharides in cultured Cordyceps militaris L.Link.
