Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ) are transcriptional coactivators that serve as downstream effectors of the Hippo signaling pathway. YAP/TAZ play a crucial role in cancer development, progression, metastasis, and drug resistance. It has been shown that YAP/TAZ are closely associated with the EGFR signaling pathway and involved in resistance to EGFR inhibitors. In addition, YAP/TAZ are frequently overexpressed and/or activated in cancer cells that are resistant to EGFR inhibitors, and inhibition of YAP/TAZ restores their sensitivity to EGFR inhibitors. This chapter presents the current knowledge of the cross talk between YAP/TAZ and EGFR signaling, the role of YAP/TAZ in resistance to EGFR inhibitors, and the mechanisms of the YAP/TAZ-mediated resistance. Moreover, the potential combinational treatment for circumventing the YAP/TAZ-mediated resistance to EGFR inhibitors is discussed.
