Abstract All myasthenic disorders result from structural or functional alterations in one or more components of the NMJ. To facilitate understanding of the mechanisms known to cause these disorders, this chapter reviews the classical anatomic features and current notions of the molecular architecture of the NMJ. The clinical, electrophysical, and pathological aspects of the individual myasthenic disorders are discussed in subsequent chapters.
