Trehalose is a widespread nonreducing disaccharide,which functions as a signaling molecular in the regulation of plant development and the response to abiotic stress.However,how it signals is still not clear.Therefore,the identification and characterization of the trehalose signaling mutants is an effective method to elucidate the trehalose signal transduction pathway.In this paper,a method to screen the trehalose signaling mutant was developed.The results showed that high concentration trehalose inhibited the plant growth,although low concentration trehalose is essential for plant growth and development.With the addition of 50 mmol·L-1 trehalose in the medium,growth of wild-type seedlings was significantly arrested.Thus 50 mmol·L-1 trehalose is suitable for screening the trehalose insensitive mutants.Further study showed that the growth inhibition effect of high concentration trehalose could be released by glucose or sucrose.Thus these metabolizable sugars could not be added to the trehalose signaling mutant screening medium.
