HPLC determination of etomidate in human plasma

Objective:To develop an HPLC method for the determination of etomidate in human plasma.Method:The Hypersil ODS C18(250 mm×4.6 mm,10 μm)was used as analytical column with a mobile phase consisted of methanol-0.062%ammonium acetate solution(60∶40).The flow rate was 1.0 mL·min-1and detection wavelength was set at 240 nm.Results:The linear ranges were 0.025-2.5 μg·mL-1 for etomidate in human plasma.The correlation coefficients were 0.9998.The etomidate low limit of detection was 12 ng·mL-1.The method recoveries were 96.8%-102.5%.The extraction recoveries were 80.7%-85.0%.Within-day and between-day RSD(n=3)were less than 5.9%.Conclusion:The method is reliable and sensitive for determination of etomidate in human plasma.
