Content Determination of Hyperoside in Different Parts of Hypericum perforatum from Different Habitats by RP-HPLC

OBJECTIVE:To develop the method for the content determination of hyperoside in Hypericum perforatum,and to determine the content of hyperoside in different parts of H.perforatum from different habitats.METHODS:RP-HPLC method was adopted.The determination was performed on Boston Breeze-C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-0.1% phosophoric acid(40:60,V/V) at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 360 nm.RESULTS:The linear rang of hypericin was 0.14～2.80 μg·mL-1(r=0.999 4) with an average recovery of 95.97%(RSD=1.16%,n=6).The contents of hypericin in the flowers and leaves of H.perforatum from Sichuan province were the highest.CONCLUSION:The method is simple,accurate and reproducible for the quality control of H.perforatum.
