Effects of Tween-80 on the Growth and Content of Isoflavones in the Pueraria lobata Hairy Roots

This article studied the effects of Tween-80 on the growth of hairy roots and the synthesis and release of second substances,which was added into the Pueraria lobata hairy roots suspension culture medium with different concentrations. The results indicated that the supplement of 2% Tween-80 was suitable,because it not only increased the content of puerarin in the hairy roots, but obviously promoted the excretion of puerarin,daidzein and total isoflavones in the medium . Contrast to the control,the contents of every substances in the medium could be raised 24.2%,50% and 46.7%,respectively. When the hairy roots were treated continuously with 2% Tween-80 for 72h,they still grew well and the fresh weight increased 1.5-fold than the control. But the effects were different on the accumulation and release of several isoflavones in the hairy roots and the culture medium. After the treatment of 48h, the content of total isoflavones in the medium was 38 times than that in the hairy roots.
