Determination and Analysis of Polysaccharide from Capparis spinosa Ripe Fruits by HPCE

OBJECTIVE To isolate polysaccharide from Capparis spinosa ripe fruits,determine the content of the polysaccharide and analyze its monosaccharide components.METHODS The contents were deternined by phenol-sulfuric acid method;HPCE was applied to analyze its monosaccharides.RESULTS The content of the coarse polysaccharide and purified polysaccharide(after getting rid of protein by sevage) were 39.95% and 66.42%,respectively.The polysaccharides were mainly composed of Rha∶Xyl∶Glu∶(Fru and Man)∶Ara∶Gal=3∶(5∶48)∶100∶21∶19.CONCLUSION The optimized way of coloration of phenol-sulfuric acid method can be easily controlled and operated;HPCE is a good way to analyze the monosaccharide components of polysaccharides.
