Objective The study was to investigate the effect of different process parameters to the yield of ethanol-soluble protein from Gnaoderma lucidum, aims to provide theoretical guidance for industry production of ethanol-soluble protein from Ganoderma lucidum. Methods Ethanol-soluble protein was extracted from Ganoderma lucidum. The effects of extraction time, extraction temperature, pH, material-liquid ratio and extraction frequency on the yield of ethanol-soluble protein were studied with single factor and orthogonal tests. And then the optimum extraction technology for ethanol-soluble protein from Ganoderma lucidum were determined. Results The results of single factor test showed that the yield of ethanol soluble protein from Ganoderma lucidum was the highest when material-liquid ratio was 1:40, extration time was 2 h. The effects of factors on yield of ethanol-soluble protein from Ganoderma lucidum from big to small in order was pH material-liquid ratio extraction temperature concentration of ethanol. The optimal condition of ethanol-soluble protein was as follows: concentration of ethanol 60%, pH 2.5, material-liquid ratio 1:40 and extration temperature 60 ℃. Conclusion Through single factor and orthogonal tests , we found an optimal condition on yield of ethanol-soluble protein from Ganoderma lucidum, which provide a theoretical guidance for industry production of ethanol-soluble protein from Ganoderma lucidum.
