Background Q-switched 1064 nm Nd:YAG laser (1064-QSNYL) was used to rejuvenate photoaged skin. In this study, we explored the effects of 1064-QSNYL on miR-27a-3p, TGFBR1, and Smad4 in photoaged rat skin. Methods Wistar rats were irradiated with ultraviolet A (UVA) and ultraviolet (UVB) for 12 weeks. Skin photoaging was verified using dermoscopy, HE staining, Sirius red staining and hydroxyproline content. Photoaged rat skin was treated with 1064-QSNYL at 2.5 J/cm2. The relationship between miR-27a-3p, TGFBR1, and Smad4 in HaCaT cells was identified by real-time PCR. The expression of miR-27a-3p was detected using real-time PCR. The expression of TGFBR1, Smad4, and Col3a1 was detected using real-time PCR, western blotting and/or IHC. Results Typical photoaging characteristics were observed in UV-irradiated rat skin. TGFBR1 and Smad4 are direct targets of miR-27a-3p in HaCaT cells. 1064-QSNYL treatment downregulated miR-27a-3p and activated the TGFBR1-Smad4-Col3a1 axis in photoaged rat skin. Conclusion 1064-QSNYL activates the TGFBR1-Smad4-Col3a1 axis via miR-27a-3p in photoaged rat skin.
