Effect of endotoxin on hepatocyte mitochondrial energy metabolism in rats and intervention of astragalus membranacenus injection

[Objective] To observe the effect of endotoxemia(ETM) on hepatocyte mitochondrial energy metabolism in rats and the intervention of astragalus membranacenus injection.[Methods] Forty healthy adult rats were randomly divided into normal control group and two LPS groups(6,12 hours after injected with LPS) and two treatment groups(treated with astragalus membranacenus injection after the model was induced 6,12 hours).Then hepatocyte mitochondria was separated to measure the activities of SDH and CCO.At the same time,the level of hepatocyte mitochondrial adenylic acid energe charge(EC) was measured by high performance liquid chromatography(HPLC).[Results] Mitochondrial SDH,CCO activities were reduced and its EC decreased after LPS was injected 6 hours,and which were more obvious after the model was made 12 hours.Compared with respective LPS groups,hepatocyte mitochondrial SDH,CCO activities and the level of EC increased obviously in astragalus membranacenus treatment groups.[Conclusion]ETM can significantly decrease the energy reserve of hepatocyte mitochondria.Astragalus membranacenus injection can increase the level of oxidative phosphoric acid and mitochondrial adenylic acid EC,ameliorate mitochondrial energy synthesization and metabolism and protect the liver cell.
