Abstract Optimal conditions for protoplast formation and regeneration in Bifidobacterium pseudolongum and Bifidobacterium longum were determined. Protoplasts were formed by lysozyme treatment together with protease or crude α-amylase showing proteolytic activity. Cell wall digestion of both strains was insufficient when the cells were treated with lysozyme alone. Regeneration of protoplasts from both strains was accomplished with a hypertonic medium containing raffinose, MgCl 2 , and CaCl 2 as stabilizers. Using optimal conditions, it was possible to prepare protoplasts of both strains and regenerate them with a frequency of 10 −3 to 10 −1 .
