Effect of TGF-β_2 on guinea pig scleral fibroblast culture in vitro

Objective To explore the technique of guinea pig scleral fibroblast (GSF) culture in vitro and to evaluate the effect of transforming growth factor-β_2(TGF-β_2) on the proliferation and collagen synthesis of GSF cultured in vitro.Methods GSFs were cultured by primary culture in vitro.Different concentrations of TGF-β_2 (0.00 ～50.00 μg·L~ -1 ) were added to the 3～6 passages of cells,and then the effect of TGF-β_2 on the proliferation and synthesis of collagen was observed using MTT method and Chlorozone T.Results TGF-β_2 could promote the proliferation and collagen synthesis of GSF cultured in vitro in a dose-dependent manner with a peaked action at the concentration of 10.00 μg·L~ -1 .Conclusion TGF-β_2 could promote the proliferation and collagen synthesis of GSF cultured in vitro.
