Objective To investigate the characteristics and its relationship with clinical features of the mutation of KRAS gene in patients with colorectal carcinomas(CRC). Methods Tumor cells were collected from paraffin-embedded tumor specimens from 151 patients with CRC, then genomic DNA was extracted and mutations of KRAS gene(codon 12 and 13) were detected by amplification refractory mutation system(ARMS). Results KRAS gene mutation was identified in 63 cases(41.7%). 6 types of mutation were detected at codon 12(53 case, 35.1%),and the most common mutation type was Gly12Asp(29 cases, 19.2%). One type of mutation(Gly13Asp) was detected in codon 13(10 case, 6.6%). No significant association was observed between KRAS gene mutation and gender, age, tumor site, degree of differentiation, type of specimen(P 0.05). Conclusions KRAS gene mutation rate was high in patients with CRC. There was no significant association between KRAS gene mutation and gender,age, tumor site, degree of differentiation, and type of specimen.
