The transcription factors NRF2 and p53 mediate the expression of many antioxidant genes in cells adapting to oxidative stress. Using three-dimensional cultures of normal and premalignant breast epithelial cells and mathematical modeling, Pereira et al. found that the oxidative stress responses of NRF2 and p53 were coordinated. Their models suggested that NRF2-mediated tolerance to oxidative stress was critical for normal duct development but, in premalignant tissue, may permit the emergence of p53 mutations that drive malignant progression and render NRF2 dispensable.
