PB2038: THE POTENTIAL MECHANISM OF TCM TREATMENT OF CHRONIC APLASTIC ANEMIA BASED ON DATA MINING AND NETWORK PHARMACOLOGY

Topic: 11. Bone marrow failure syndromes incl. PNH - Biology & Translational Research Background: Aplastic anemia is a disease characterized by anemia, hemorrhage and infection due to low hematopoietic function of bone marrow, which is divided into acute aplastic anemia and chronic aplastic anemia. For chronic aplastic anemia, the disease progresses slowly and the symptoms are relatively mild. The combination of cyclosporine and androgen is often used in modern medicine to treat chronic aplastic anemia according to the severity of the disease. Especially in China, the treatment of aplastic anemia with traditional Chinese medicine is a widely used treatment method. For patients with obvious changes in blood items, the effective rate is high, and some patients can even use Chinese medicine alone to obtain better curative effect. The effective components and mechanism of action of traditional Chinese medicine are relatively complex. Therefore, we collect the traditional Chinese medicine with high frequency of use in the literature to predict the potential effective targets through data mining. Aims: To explore the potential mechanism of TCM treatment of chronic aplastic anemia. Methods: A total of 377 articles on the treatment of aplastic anemia by traditional Chinese medicine from January 1, 2012 to January 1, 2022 were collected through the document platform of CNKI,The most frequently used Chinese medicine was calculated and obtained.The TCMSP, GeneCards and STRING databases are used to obtain the drug components, action targets and disease targets of traditional Chinese medicine and the intersection targets of traditional Chinese medicine and disease. The DAVID and microbiological information databases are used to carry out GO and KEGG analysis on the action targets of traditional Chinese medicine in treating AA. Results: According to the literature data mining, the first six representative traditional Chinese medicines for the treatment of aplastic anemia were used from high to low: 207 times of astragalus, 197 times of angelica, 158 times of dodder seed, 151 times of rehmannia, 150 times of ligustrum lucidum, 147 times of atractylodes macrocephala. After screening and weight removal, 33 Chinese herbal components and corresponding 203 target targets were obtained, and 74 targets related to aplastic anemia. GO enrichment analysis showed that the most relevant biological processes (BP) were positive regulation of gene expression, negative regulation of apoptosis process, and positive regulation of transcription; The cell components (CC) have the strongest correlation with extracellular space, extracellular area, cell surface, etc; The strongest correlation of molecular function (MF) is enzyme binding, cytokine activity, protein binding, etc. The most relevant KEGG enrichment analysis includes cancer pathway, lipid and atherosclerosis, fluid shear stress and atherosclerosis, the role of AGE-RAGE signaling pathway in the complications of diabetes, prostate cancer and other pathways. Summary/Conclusion: The above six traditional Chinese medicines can treat aplastic anemia through the above multiple mechanisms.
