Objective The different doses of UVA1 were screened to investigate their influences to the morphology and cell activity of the dermal of artificial skin in order to provide a theoretical basis of dermal of artificial skin photo-damage model. Methods After different doses of UVA1 exposure(0J/cm2-80J/cm2),the number of changes of dermal fibroblasts was observed from the morphology by histology staining,and the dermal fibroblast activity was measured by the MTT assay. Results Histology staining showed that major alterations of the number of dermis fibroblasts were observed within the superficial part of dermal equivalent,and inversely with the increase of the UVA1 dose. MTT assay showed that there was no obvious depression of fibroblast activity in group 20J/cm2 and group 30J/cm2 (P0.05)after UVA1 exposure. Fibroblast activity was decreased obviously (P0.05,P0.001)when the dose was more than 40 J/cm2 ,and this change appeared dose-dependent. Conclusion The dose of UVA1 irradiation which was higher than 40J/cm2 was orginal dose leading to the dermal fibroblast photo-damage of artificial skin.
