Objective To study the effect of HIF-1α,VEGF,p53 and CD31 on vasculogenic mimicry in the melanoma transplant tumour.Methods The mouse melanoma transplant tumor model was made with B16 melanoma cell suspending liquid.When tumor appears,one mouse was chosen each day randomly to execute.114 cases tumor specimens were obtained for 12 days totally to conduct HE stain,immunohistochemistry stain for HIF-1α,VEGF,p53 and CD31 and CD31-PAS double stain.Results The vasculogenic mimicry and endothelial-depended vessel co-exist in the tumor tissue at the early phase of melanoma;From day 9 to day 12,this structure was replaced by endothelial-depended vessel.The number of CD31,HIF-1α positive cell around vasculogenic mimicry were more than that of far away vasculogenic mimicry.The ratio of CD31 positive cell around endothelial-depended vessel had significant difference with that of far away from it.The number of VEGF,p53 positive cell around vasculogenic mimicry were more than that of around endothelialdepended vessel.There were correlation between vasculogenic mimicry density and the number of CD31,HIF-1α,VEGF positive cells around vasculogenic mimicry. The density of endothelial vessel had correlation with the number of CD31 positive cell around endothelial-depended vessel.Conclusions The vasculogenic mimicry is one sort of temporary blood-providing pattern.The HIF-1α,VEGF,p53,CD31 all take part in the development of vasculogenic mimicry and CD31,HIF-1α play key role in this procession.
