摘要
Wheat sharp eyespot caused by Rhizoctonia cerealis Van der Hoeven,is a soil-borne disease,which has significantly threatened the high and stable production of wheat in China.In order to study the pathogenesis of R.cerealis,the enzyme,the enzymolysis time and the osmotic stabilizer for protoplast preparation were optimized.Two means the PEGprotoplast and Agrobacterium tumefaciens mediation were emloyed to transform R.cerealis.An efficient and stable system of protoplast preparation for R.cerealis was developed,and there was no difference between regenerated strains and original strain in morphology,speed of growth and pathogenicity.No transformant was obtained by both methods.The protoplast preparation system for R.cerealis provided a basis for genetic transformation,protoplast fusion and so on.
