Purpose To study the effects of prevention and treatment in postmenopausal osteoporosis with different dose hormone replacement therapy. Methods Sixty-nine postmenopausal women were divided into three groups(n=23 per group):Group A to receive daily oral conjugated estrogen(Premarin 0.625 mg) in combination with medroxyprogesterone acetate and calcium carbonate(1 g/d);group B to receive daily oral conjugated estrogen(Premarin 0.3 mg)in combination with medroxyprogesterone acetate and calcium carbonate(1 g/d);group C to receive placebo and calcium carbonate(1 g/d) for two years.We observed bone mineral density,biochemical markers and LI?6.We also followed up endometrium thickness and breast.Examinations were scheduled to be performed pre-therapy and after 6,12,18,24 months of therapy. Results Bone mineral density of group A and B increased significantly one year after treatment.Bone mineral density of group C decreased in one year.No endometrial hyperplasia was observed in any of the three groups.The biochemical markers decreased at 6 months and remained low thereafter.We also observed a similar decrease in serum levels of IL?6. Conclusions Lower-dose HRT are effective in long-term prevention of postmenopausal osteoporosis,and prevents endometrium hyperplasia and decrease the vaginal bleeding rate.
