摘要
Psoriasis is a chronic skin disease with severe consequences for the whole organism. Therefore, the lack of knowledge of the factors predisposing an individual to the psoriasis development, leads to investigate changes in the plasma proteomic profile of psoriatic patients. A proteomics approach was used to analyze the expression of proteins in plasma from psoriatic patients and healthy controls. The analysis was performed using gel electrophoresis, followed by LC-MS/MS (Q-Exactive OrbiTrap). Obtained data showed a decrease in the level of proteins involved in lipid metabolism in psoriatic patients’ plasma. These changes were accompanied by the increased expression of proinflammatory and signaling molecules - particularly transcriptional factors regulating cellular proliferation. In addition, the lipid peroxidation product - HNE generates higher level of adducts with proteins with catalytic activity in the plasma of psoriatic patients. The results presented herein identify several new proteins that are involved in signal transduction and lipid metabolism, which changes in expression are associated with psoriasis development, therefore this knowledge may help to the design of more specific pharmacotherapy.
