摘要
Coumarin-based metal complexes have emerged as promising multifunctional agents exhibiting a broad spectrum of pharmacological activities, notably anticancer, antibacterial, and antioxidant effects. The coumarin scaffold offers significant structural versatility and strong metalchelating properties, which collectively enhance the therapeutic potential of these complexes. In the field of oncology, coumarin-metal conjugates display notable cytotoxicity by inducing apoptosis and modulating intracellular Reactive Oxygen Species (ROS) levels. These complexes also exhibit strong antimicrobial activity, primarily by disrupting microbial membranes and inhibiting key bacterial enzymes, suggesting potential efficacy against drug-resistant pathogens. Additionally, their antioxidant activity stems from their capacity to scavenge free radicals and chelate redox-active metal ions, effectively reducing oxidative stress. Among these, coumarin-Schiff base metal complexes are particularly significant due to their high DNA-binding affinity, which reinforces their potential as chemotherapeutic and antimicrobial agents. The inclusion of transition metals-such as copper, zinc, iron, and ruthenium-enhances the physicochemical stability of the complexes and plays a critical role in modulating their biological activity. This mini-review highlights recent advances in the design and development of coumarin-based metal complexes, with emphasis on their mechanisms of action and therapeutic applications. The pharmacological contributions of various transition metals are discussed to provide insight into the potential of these compounds as next-generation multifunctional drug candidates.
