Five different methodologies have been used to load miRNAs into sEVs: transfection by Exo-Fect or cholesterol-modified miRNA and membrane permeabilization by a detergent (saponin), electroporation or heat shock (1). The modified sEVs were then characterized regarding their loading efficiency by fluorescence and qRT-PCR analyses (2), bioactivity in the HEK-293T reporter cell line (3), cell toxicity using a cell viability assay (4) and capacity to transfect cells (5).
