Effect of expression results on mRNA in situ hybridization using different decalcification methods in rabbit bone growth plate cells

Objective To explore the effect of expression results on mRNA in situ hybridization using different decalcification methods in rabbit bone tissue.Methods The preparation of sense and anti sense mRNA probes were made by the rabbit Col Ⅱ genes and used in situ hybridization in the rabbit bone tissue.Results The best decalcification method is the 10% EDTA + ATM solution between the several different decalcification methods.Conclusion Using the 10% EDTA + ATM solution can inhibit the RNAase totally and provide the good expression results in situ hybridization in the rabbit bone growth plates.
