Abstract Purpose To explore suzetrigine, a novel voltage-gated sodium channel inhibitor for pain management. Summary Suzetrigine is a first-in-class, oral peripheral sodium channel inhibitor that selectively targets Nav1.8 channels and was recently approved for the treatment of moderate to severe acute pain. In phase 3 trials, suzetrigine demonstrated superior efficacy to placebo in reducing pain intensity after surgery and was generally well tolerated. Studies for nonacute pain indications, including diabetic peripheral neuropathy and painful lumbosacral radiculopathy, are in progress. Conclusion Suzetrigine represents an advancement in pain management as a nonopioid alternative for pain with potentially lower abuse risk and potentially fewer serious adverse effects when compared to opioids. While initial results are promising, further research is needed to establish its long-term safety and efficacy for various painful conditions. Research will continue in the area of analgesics that selectivity inhibit peripheral sodium channels.
