Based on the tertiary structure of protein, contact number categories from a database of two and multi-state proteins is created.A linear regression model is built for predicting the protein folding rate using protein contact number. The correlation between predicted folding rates and experimental folding rates is 0.832 with Cα atom as characteristic. With N atom as characteristic, the correlation with folding rates reaches 0.903.It is proved that the residues interactions in a protein are an important determinant of the protein folding rate. The interaction effect of N atom and other atoms on the folding rate is superior to the interaction effect of Cα atom and other atoms.
