The experiment study of the effect of Xianlinggubao medicated serum on the co-culture system of mouse osteoblast and osteoclast

Objective To observe the effect on mouse osteoblast and osteoclast in co-culture system by different concentrations of Xianlinggubao medicated serum.Methods The co-culture system of the mouse osteoblast MC3T3-E1 and osteoclast induced by mouse single nucleus macrophage RAW 264.7 were established.The experiment groups were consisted of different concentrations of Xianlinggubao medicated serum and blank control group.The osteoblast mineralization tubercle and osteoclast bone absorption lacuna number were observed by light microscope and the the level of ALP activity in the nutrient fluid was tested by PNPP method.The co-culture supernatant fluid in 1,3 day were collected and the contents of OPG,RANKL were measured by ELISA.Results 7 days later,the number of mineralized tubercle of high concentration group was higher than that of other two groups obviously(P0.01),and it was higher in low concentration group than that of control group(P0.05).7 days later,the count of mineralized nodus of control group was higher than that of other two groups obviously(P0.01),and there was no significant difference between other two groups.1 day later,the ratio of OPG/RANKL of high and low concentration groups were higher than that of control group(P0.05,P0.01),3 days later the ratio in high concentration group was higher than that of the other two groups(P0.05,P0.01),and the ratio in low concentration group was higher than that of control group(P0.05).Conclusions Xianlinggubao medicated serum could promote the ability to produce mineralized nodus of the osteoblasts in co-culture system and reduce the count of absorption lacuna on bone chips.Xianlinggubao medicated serum could increase the ratio of OPG/RANKL and present time and dosage dependence.
