BMP2 Induces Adipogenic Differentiation of C3H10T1/2 Cells

To investigate the role of bone morphogenetic protein 2(BMP2)in inducing the adipogenic differentiation from mesenchymal cell and exploit the potential applications for the treatment of lipometabolism dysfunctions,the mouse multi-potent C3H10T1/2 stem cells were treated with 20 μg/ml BMP2 for various periods.The adipogenic process was characterized with Oil Red O staining in BMP2 treated cells.RT-PCR was employed to detect the existence of key molecules along the BMP signaling pathway,including BMPRⅠ,BMPRⅡ and Smad1/5/8.Western blot assays were used to examine the levels of phosphorylated Smad proteins and p38.The levels of an adipogenic marker,aP2 and transcription factors PPARγ,C/EBPα and C/EBPβ,were detected by quantitative RT-PCR.After the induction of BMP2,the Oil Red O demonstrated an obvious induction of adipogenic differentiation in C3H10T1/2 cells.The increased phosphorylation of Smad proteins and p38 were also observed.The levels of PPARγ,C/EBPα,C/EBPβ and aP2 were upregulated at various degrees.The results demonstrated that BMP2 exhibited a potency to induce the adipogenic differentiation in C3H10T1/2 cells in a time-dependent manner.
