摘要
Anxiety disorders, the most prevalent mental health conditions with a lifetime prevalence of up to 28%, represent a major global health challenge due to inadequate treatment options. 1 Emerging research highlights the crucial role of lipid metabolism dysregulation in anxiety pathophysiology. Of particular interest is elongase of very long-chain fatty acids 6 (Elovl6), the key enzyme responsible for C12-C18 fatty acid elongation. This enzyme shows particularly high expression in the human cerebral cortex, a region vital for emotional regulation. Accumulating evidence suggests that dysregulation of Elovl6 contributes to the pathogenesis of neuropsychiatric disorders. A large-scale case-control study demonstrated that reduced serum Elovl6 activity is significantly associated with an increased risk of suicide. 2 Moreover, transcriptomic analyses have identified Elovl6 as one of the genes most responsive to fluoxetine, 3 a commonly prescribed antidepressant. These findings strongly implicate Elovl6 in the pathophysiology of anxiety and depression, although its precise molecular mechanisms remain to be fully elucidated.
