Objective To establish an experimental model of severe acute pancreatitis induced by retrograde cholangiopancreatic duct infusion of bile salt. Methods Severe acute pancreatitis(SAP) was induced by retrograde cholangiopancreatic infusion of sodium taurocholate in 8 rats, with a sham operation group(n=8) used as control. The concentration of serum calcium, the activity of serum amylase were measured. The histological appearances of the pancreas were also observed. Results Compared with the sham operation group, the activity of serum amylase and the mortality were increased. Serum calcium was decreased in SAP group. Pancreatic ledema, haemorrhage, acinar cell necrosis and extensive fat necrosis were found in the pancreas of SAP group. Conclusion The experimental model of severe acute pancreatitis induced by retrograde cholangiopancreatic duct infusion of bile salt can be a useful tool to investigate the pathogenesis and treatment of severe acute pancreatitis.
