Colorimetric detection and quantification of glutathione in human serum using the peroxidase-like activity of MOF-808FA frameworks

The measurement of GSH plays a crucial role in the timely diagnosis, treatment, and overall management of many diseases, significantly impacting human health. In this research, MOF-808FA has been employed for the first time as a peroxidase-like material for the colorimetric detection and quantification of GSH. First, MOF-808-FA was synthesized using the solvothermal method and characterized through FT-IR (Fourier Transform Infrared Spectroscopy), XRD (X-ray diffraction), and SEM (Scanning Electron Microscope) techniques. The MOF’s peroxidase activity was assessed using a TMB + H 2 O 2 system. Measurement performance was accomplished in optimal pH, MOF-808FA amount, TMB concentration, and Time. Afterwards, a calibration curve was provided. Accordingly, real sample analysis and selectivity assay were performed. MOF-808FA was synthesized and characterized successfully. The results of the analysis assay showed that the method exhibited a linear range of 0–152.7 μM, with a detection limit of 0.74 μM. Additionally, this method demonstrated excellent anti-interference properties. It also yielded favorable results in measuring GSH levels in human serum, with recoveries calculated to be between 104.67 % and 110.57 %. The relative standard deviation (RSD) was between 1.0 % and 2.4 %. In conclusion, MOF-808-FA demonstrated significant peroxidase activity and facilitated the development of a sensitive and selective colorimetric method for determining GSH. The utilization of MOF-808FA highlights the promising direction of enzyme mimics in colorimetric assays.
