Content determination of clobetasol propionate in Clobetasol Propionate Urea Cream by HPLC

Objective: To establish a HPLC method for the content determination of clobetasol propionate in Clobetasol Propionate Urea Cream. Methods: The analytical column was SHIMADZU VP-ODS C18 column(4.6 mm×150 mm, 5 μm) with a mobile phase consisted of 0.05 mol/L disodium hydrogen phosphate solution(85% phosphoric acid solution with pH adjustment to 2.5)-acetonitrile-methanol(42.5∶47.5∶10) at the flow rate of 1.0 ml/min, the detection wavelength was set at 254 nm. Results: The linear range of clobetasol propionate was 5-40 μg/ml, r=0.999 9, the average recovery was 98.05% with a RSD of 0.72%. Conclusion: The method is accurate, reliable, simple and feasible, and can be used in the content determination of clobetasol propionate in Clobetasol Propionate Urea Cream.
