Antibody-directed enzyme prodrug therapy (ADEPT) was designed to generate high concentration of a cytotoxic agent selectively within tumors. Preclinical studies and clinical trials have improved our knowledge of the essential requirements for a successful ADEPT system. As a potential nontoxic therapy, ADEPT may be developed as a combined therapy with other clinically useful agents.
