Molecular Classification of Diffuse Large B Cell Lymphoma

Diffuse large B cell lymphoma (DLBCL) is a heterogeneous disease with varied clinical presentation, genetic findings, response to therapy, and long-term prognosis despite possessing a uniform histologic appearance. By using a gene expression–based classifier, a high-risk subtype was further designated a molecular high-grade or double-hit signature with distinct molecular features and clinical outcomes that increases the size of the double-hit group with poor prognosis. In this review, we explore the current molecular understanding of the DLBCL subtypes and discuss the relationship with prognosis and promising targeted approaches for each associated subtype.
