Effects of GM-1 on the proliferation and differentiation of hippocampal neural stem cells in newborn rats

Objective:To observe the effects of different dose of GM-1 on the proliferation and differentiation of the hippocampal neural stem ca lls. Methods: The neural stem cells isolated from the hippocampus of the newborn rats were cultured in the medium(negative-control group: DMEM/F12 , B27 and glutamate, pH=7.0), the medium containing bFGF, and the medium contain ing different dosage of GM-1, respectively. The proliferation of different group s was examined by counting the number of neurospheres and the differentiation of neural stem cells was identified by the immuncytochemical technique. Re sults: There was no significant difference in the number of neurospheres b etween three experimental groups and the positive-control group, but the differe nce between experimental groups and the negative-control group was significant. Conclusion: GM-1 has effects on the proliferation of the neural stem cells, while it has no obvious influence on the differentiation of the neural stem cells.
