Objective To develop selenium-deficient Kunming mice by feeding with self-made selenium-deficient diet and observe the effects of selenium-deficiency on Kunming mice myocardium. Methods A selenium-deficient mouse food was prepared with local yeast.Young Kunming mice were fed with the selenium-deficent diet for over 4 months.Selenium concentrations in the blood,liver,and myocardial tissue of the mice fed with selenium-deficient diet were determinated.The myocardial enzymes in the treated mice were examined.The ultrastructures of the treated mice myocardial tissues were also observed. Results The selenium concentration in the mouse food prepared with local yeast was 0.016mg/kg and the standard of selenium-deficiency.The selenium levels in the serum,liver,and myocardial tissue of Balb/c mice fed with the selenium-deficient diet for 4 months were 0.241mg/kg,0.219mg/kg,and 0.187mg/kg.Swelling mitochondria and heteromorphic nucleus were observed in the selenium-deficient cardiomyocytes.The level of myocardial enzymes in the sera of the selenium-deficient mice increased. Conclusions A selenium-deficient mouse diet was successfully developed with local yeast,Selenium-deficient mouse can be prepared by feeding this diet.Selenium-deficiency mainly cause injuries in myocardial mitochondria of the Kunming mice.
