Metalloproteins play key roles in many of the most unique and challenging chemical transformations in nature, however, our knowledge of them is still limited. Metalloprotein design provides a deeper understanding of biochemical principles that govern these systems and allows chemists to engineer better catalysts. Critical to the advancement of this field is the conception and development of novel strategies for protein design and engineering. Here, we describe major advances in those design tools and strategies, how they have been implemented, insights they have provided into the bio-coordination chemistry of metalloproteins, and the future directions they are leading the field.
