摘要
To express and serologically evaluate the protective antigen Eg95 recombinant protein of Echinococcus granlosus for the purpose of usage in prevention or treatment in cystic echinococcosis(CE),the prokaryotic expression vector pET28a-Eg05 was constructed and the constructed plasmid was transferred into E.coli BL21(DE3)with induction of IPTG for expression.The recombinant proteins thus obtained were purified with His-binding agarose by affinity chromatography.Eleven samples of sera taken from patients with CE were used to evaluate the serological reaction of Eg95 recombinant protein.It was found that the expressed pET28-Eg95 recombinant protein was detected as a band of 15 kDa by SDS-PAGE assay.As demonstrated by Western blotting,it could react specifically with serum samples of patients with CE.In the serological reaction of this recombinant protein with 11 serum samples of patients with CE,8 serum samples showed positive reaction.From these observations,it is evident this recombinant protein may be considered as a protective antigen in CE patients and would be useful for the treatment of CE infection.
