Optimization of Factors Related to High Frequency Plant Regeneration of Pueraria lobata

The effects of several factors on dedifferentiation and redifferentiation of Pueraria lobata earthnut were studied.The results showed that the best disinfection way of Pueraria lobata stems with a bud was firstly treated with 70% ethyl alcohol for 30 seconds then treated with 0.1% HgCl2 for 15 minutes,and the optimum regeneration culture medium was MS+NAA 0.5mg/L + 6-BA 5.0mg/L.Liquid culture with sucrose 30g/L,pH 6.0,cultured in light at 25℃ gave a best result for high frequency plant regeneration of Pueraria lobata.
