Determination of Bupivacaine in Rat Myocardium by LC-MS/MS

A LC-MS/MS method was established for the determination of bupivacaine in rat myocardium.A C 18 column was used with the mobile phase of methanol-0.1%formic acid solution(55∶45)and lidocaine as the internal standard.Electrospray ionization(ESI)source was operated in positive ion mode and multiple-reaction monitoring (MRM)mode.The detected ions were m/z 289→140 for bupivacaine and m/z 285→86 for the internal standard.The calibration curve for bupivacaine was linear in the range of 0.01-20.0μg/g in myocardium.The recovery was 66.45%74.47%with the intra-and inter-day RSDs less than 7%.
