Objective To investigate the effect and the mechanisms of Telmisartan on the expression of klotho protein in the brain choroid plexus in mice.Methods 32 male Kunming mice were divided randomly into four groups: the normal control group,NOS blockade(L-NAME) group,Telmisartan group and L-NAME plus Telmisartan group.All animals were administered by corresponding medicine for four weeks,respectively.The expression of klotho protein in the brain choroid plexus were measured by immunohistochemistry and Western blot technigues.The change of expression of RhoA/ ROCK mRNA were detected by RT-PCR.Results Compared with the control group,the expression of RhoA and Rock mRNA increased significantly(P0.01) and the expression of klotho protein decreased sharply(P0.01) in the L-NAME group.Telmisartan reversed these changes(P0.01).Conclusion These findings suggest that Telmisartan could intervene down-regulate the expressionof klotho protein induced by L-NAME.The mechanism might be related to inhibit the activities of Rho / ROCK signaling pathway.
