Objective To establish the spectrophotofluorimetry of determination the content of salidroside. Methods The optimum fluorescence excitation wavelength and emission wavelength was investigated. The effects of laying time and temperature were investigated and the optimum analytical parameters were obtained. Results The maximum ex- citation wavelength of salidroside was λ ex = 273 nm and the maximum emission wavelength was λ em = 301 nm. The lin- ear range of salidroside was 0. 01 ~ 0. 10 mg / L with the linearly dependent coefficient of 0. 999 0. The Detection Limit( DL) was 0. 005 mg/L and the Relative Standard Deviation( RSD) was 0. 32% for 5 independent determinations of 0. 05 mg / L salidroside solution. The content of salidroside in Rhodiola crenulata( from Xizang,China) was extracted and determined using the founded method. Conclusion The spectrophotofluorimetry was sensitive and rapid with a low detection limit. The method can be used to determine the content of salidroside in crude drugs.
