Abstract The fluorescence spectra of the synthesized C5-substituted pyrimidine nucleosides with polycyclic aromatic substituents indicated that they were sensitive to environmental factors. The fluorescence of oligonucleotides containing these nucleosides was measured when double-stranded DNA was formed. When the oligonucleotide containing the nucleoside with a 2-phenanthryl group was hybridized with a fully matched sequence, the fluorescence intensity was 17 times higher than that when 2-phenanthryl-modified oligonucleotide was hybridized with a mismatched sequence.
