Aim:To explore the efficacy of p53 gene therapy combined with radiotherapy against human ovarian cancer cells. Methods:Human wild-type p53 gene eukaryotic expression vector and the same vector without p53 gene were introduced into SKOV-3 cells respectively by lipofectin-mediated gene transfection. The expression of p53 gene was detected by Northern blot and Western blot. The colony formation of transfected cells was observed after irradiating with X-rays.Results:The exogenous p53 gene was effectively expressed in the transfected cells. After irradiating with X-rays, the colony number of cells transfected with p53 gene was much lower than that of transfected and untransfected control. Conclusion:Exogenous wild-type p53 gene can enhance the sensitivity of human ovarian cancer cells to radiotherapy. The combination of p53 gene therapy and radiotherapy may be more effective in ovarian cancer treatment.
